Prussian Blue Reaction according to Perls

Procedure:

1. Stain in Potassium Ferrocyanide solution at 60°C for 15 minutes. Filter solution after heating and before use.
2. Wash in distilled water.
3. Stain in Safranin O for 2 - 5 minutes.
4. Wash in 1% acetic acid.
5. Dehydrate in 96% alcohol two times then 100% alcohol.
6. Clear in xylene and mount.

Results:

Nuclei - red
Hemosiderin - blue/green

Solutions:

A. Potassium Ferrocyanide Solution

• Potassium Ferrocyanide - 1 gm
• Distilled Water - 50 ml
• 2% Hydrochloric Acid - 50 ml

B. Safranin O

• Safranin O (C.I.#50240) - 0.2 gm
• 1% Acetic Acid - 100 ml
